响应面优化双酶法提取云南滇黄精水不溶性膳食纤维工艺

Process Optimization of Double Enzymatic Extraction Technology ofInsoluble Dietary Fiber from Polygonatum Kingianum byResponse Surface Methodology

摘要: 为促进滇黄精渣的综合利用,以云南滇黄精药渣为原料,利用淀粉酶和蛋白酶提取药渣中的水不溶性膳食纤维(IDF)。通过单因素试验,研究加酶量、pH 和温度对黄精 IDF 提取率的影响:采用 Box-Behnken 响应面分析法优化确定提取滇黄精 IDF 的工艺条件。结果显示:滇黄精水不溶性腾食纤维的最优提取工艺为 -淀粉酶的添加量176U/g、酶解pH 6、酶解温度56C;碱性蛋白酶的添加量6100 U/mg、酶解 pH 9、酶解温度55C。在此条件下,滇黄精IDF 平均提取率可达62.86%,IDF 的持水力为9.76g/g,持油力为5.74g/,膨胀力为5.86 ml/g。该工艺条件稳定可靠,可为提升滇黄精副产物的附加值及开发保健食品提供参考。

Abstract: In order to promote the comprehensive utilization of Polygonatum kingianum residue , the insol-uble dietary fiber (IDF) from Polygonatum kingianum residue in Yunnan Province was extracted by am.ylase and protease. The effects of the amount of enzyme , pH and temperature on the extraction rate of dietary fiber were studied by single factor experiment. The Box-Behnken response surface analysis was usedto optimize the extraction conditions of dietary fier from Polygonatum kingianum residue. The resultsshowed: the a-amylase addition amount was 176 U/g, the enzymatic hydrolysis pH was 6, the enzymatichydrolysis temperature was 56 C ; the alkaline protease addition amount was 6 100 U/mg, the enzymatichydrolysis pH was 9, the enzymatic hydrolysis temperature was 55 C . Under this condition , the averageextraction yield of IDF was 62. 86% , the water holding capacity of IDF was 9. 76 g/g, the oil holding capacity was 5.74 g/g, and the expansion capacity was 5. 86 ml/g. The extraction process was stable andreliable, which provided a reference forimproving the value of Polygonatum kingianum by product and de.veloping of health food.
